Proposed study involves developing nanobioconjugate based on Poly (D,L-lactide-co-glycolide) (PLGA) coated with chitosan. The nanobioconjugate carries anti-EGFR Monoclonal Antibody (mAb) to target TNBCs, anti-mouse Transferrin Receptor (TfR) antibody for drug delivery through the host endothelial system and siRNA (small interfering RNA) to specifically silence TNBCS overexpressed gene mTOR (Mammalian target of rapamycin) and to improve the anticancer potential of Paclitaxel (PTX). Methodology: siRNA-PTX loaded nanobioconjugated-mAb targeting TNBCs will be formulated and characterized. In vitro studies will be performed using TNBC and spheroid cells. EGFR, mTOR expression at mRNA and protein levels will be quantified using real time PCR and western blotting, respectively. Further assessment of cell cycle events on TNBC and spheroid cells after transfection will be investigated using flow cytometry. Subsequently, in vivo studies will be done using athymic mice model to confirm the targeting ability of the novel siRNA-PTX loaded nanobioconjugated-mAb. Outcome/Output: We aim to develop siRNA-PTXloaded nanobioconjugated-mAb specifically targeting TNBCs. Cancer statistics show that among all cancer cases in Malaysia, breast cancer is most prevalent. Hence our idea of developing siRNA loaded nanobioconjugated-mAb has tremendous therapeutic potential in Malaysia. This approach can also be used for targeted chemotherapy in breast cancer treatment.
